Pfizer’s late-stage investigational medicines for breast cancer

Atirmociclib – a highly potent CDK4 inhibitor 

  • Oral tablet taken twice daily
  • Potent CDK4 inhibitor with minimal effect on CDK6
  • May allow higher dosing with fewer side effects
  • Early data shows promise in patients who progressed on previous treatments
  • Being tested in combination with PF-07104091 (CDK2 inhibitor) to overcome resistance 

Disitamab vedotin – an antibody-drug conjugate (ADC)  

  • Given as an IV infusion once every 2 weeks
  • Novel HER2-directed ADC
  • Delivers a microtubule inhibitor (MMAE) to tumor cells
  • Shown encouraging monotherapy activity in late-line HER2-expressing mBC 

Prifetrastat – a KAT6A inhibitor  

  • Oral tablet taken twice daily
  • Blocks KAT6A and KAT6B enzymes
  • Shown promise when combined with fulvestrant to overcome endocrine resistance
