Pfizer’s late-stage investigational medicines for breast cancer
Atirmociclib – a highly potent CDK4 inhibitor
- Oral tablet taken twice daily
- Potent CDK4 inhibitor with minimal effect on CDK6
- May allow higher dosing with fewer side effects
- Early data shows promise in patients who progressed on previous treatments
- Being tested in combination with PF-07104091 (CDK2 inhibitor) to overcome resistance
Disitamab vedotin – an antibody-drug conjugate (ADC)
- Given as an IV infusion once every 2 weeks
- Novel HER2-directed ADC
- Delivers a microtubule inhibitor (MMAE) to tumor cells
- Shown encouraging monotherapy activity in late-line HER2-expressing mBC
Prifetrastat – a KAT6A inhibitor
- Oral tablet taken twice daily
- Blocks KAT6A and KAT6B enzymes
- Shown promise when combined with fulvestrant to overcome endocrine resistance
